Abstract:
With the aim of better understanding the initial defects in the anterior cavefish neural plate, we compared the size, shape and organisation of the eyefield with respect to surrounding tissues between surface fish and cavefish embryos. 3D shape and volume of the anterior neural plate was obtained using confocal Zic1 fluorescent in situ hybridization stainings at the tailbud stage. The Zic1-positive region encompassing the presumptive eye and telencephalon territories was slightly smaller in the cavefish at that stage and presented a different shape. In addition, the volume between the two ventral median sides of the eyefield was wider/higher in cavefish, suggesting a complementary pattern with the underlying posterior prechordal plate. We are now using multiplex fluorescent in situ hybridization using different markers of the eyefield and telencephalon and 3D renditions on confocal stacks to map and assign the specification defects in the anterior neural plate in the cavefish. This approach will allow us to better understand the origin of the cavefish coloboma-like phenotype.
