Abstract:
Cryopreservation is the process of storing cells, tissues, or embryos at very low temperatures for future use. This protocol reviews the process of freezing Astyanax mexicanus sperm to maintain viability when needed at a future timepoint. There are many benefits to maintaining lines in a cryopreserved state, including critical backups in case of catastrophic colony loss. Cryopreservation also allows a more efficient colony management through the improved use of space and resources within the facility. To obtain high-quality material, animals need to be in optimal health. This protocol describes the basic care required to ensure healthy fish for sample collection, in addition to the methodology for preservation, quality analysis, and subsequent fertilization.
